Selank: Tuftsin Analogue Pharmacology and Provenance

Selank is a synthetic heptapeptide built from tuftsin, an immunologically active tetrapeptide fragment of IgG heavy chain, extended with a proline-glycine-proline sequence to slow degradation. Around one hundred and forty results are indexed. As with several compounds in this catalogue, the dominant feature of the literature is where it comes from: a large share originates from Russian research institutions and appears in journals with limited international circulation.
Derivation from tuftsin
Tuftsin is Thr-Lys-Pro-Arg, a fragment of the IgG heavy chain with documented effects on phagocyte activity. Siebert et al. (Curr Med Chem, 2017) review tuftsin properties and analogues and are the best orientation point for the parent compound.
The C-terminal Pro-Gly-Pro extension in Selank is a stabilising modification. The same design appears in Semax, which is built on a different parent peptide with the same extension, and the shared motif is why the two compounds are frequently studied together.
Reported mechanisms
Volkova et al. (Front Pharmacol, 2016) report that Selank administration affects expression of genes involved in GABAergic neurotransmission. This is the most frequently cited mechanistic claim and is consistent with the anxiolytic framing in the wider literature.
A second strand concerns inflammatory gene expression. Kolomin et al. published in Regulatory Peptides (2011) on expression of inflammation-related genes in mouse spleen, and in Molecular Immunology (2014) on the temporal dynamics of that response. These connect Selank back to tuftsin's immunological origin.
Konstantinopolsky et al. (Bull Exp Biol Med, 2022) report attenuation of aversive signs of morphine withdrawal in rats. Panikratova et al. (Dokl Biol Sci, 2020) apply a functional connectomic approach to Selank and Semax effects, which is among the few studies attempting a systems-level readout.

A caution specific to this compound: a literature search on "Selank" returns results that have nothing to do with it, including papers on dental restorations and inorganic chemistry, because of author name and abbreviation collisions. Several such entries appear in the indexed set for this compound.
Any citation count for Selank taken at face value will be inflated. Filter by reading titles before drawing conclusions about the size of this literature.
Evidence assessment
Evidence base: modest and concentrated in provenance, with gene expression as the primary readout rather than behavioural or clinical endpoints. Search results require manual filtering because of name collisions.
