DSIP: Delta Sleep-Inducing Peptide and an Unresolved Forty-Year Literature

DSIP is a nonapeptide first isolated from rabbit cerebral venous blood during sleep induction experiments in the 1970s. Its literature is unusual in this catalogue: reasonably large, largely concentrated in the 1980s, and explicitly unresolved. A 2006 review in the Journal of Neurochemistry is titled "a still unresolved riddle," which is a fair summary of the position two decades later.
Origin and the naming caution
The peptide was named for the experimental context in which it was found rather than for a demonstrated mechanism. That is worth holding onto, because the name asserts a function that the subsequent literature did not cleanly establish.
Schneider-Helmert and Schoenenberger (Experientia, 1981) reported effects of synthetic DSIP on disturbed human sleep. Susić et al. (Brain Res, 1987) examined wakefulness and sleep patterns in the cat. The results across species and laboratories have not converged into an accepted mechanism.
Reported activities beyond sleep
Much of the literature concerns activities unrelated to sleep. Nakamura et al. (Eur J Pharmacol, 1988) report antinociceptive effects following central administration. Sahu and Kalra (Life Sci, 1987) report stimulation of luteinising hormone release in steroid-primed ovariectomised rats. Bjartell et al. (Peptides, 1989) describe DSIP-like immunoreactivity in gut tissue coexisting with known peptide hormones.
The breadth of reported activity without a unifying receptor is itself the central problem. No specific DSIP receptor has been convincingly identified, which is the main reason the mechanism remains open.
How to read this literature
Graf and Kastin reviewed the field twice, in Neuroscience and Biobehavioral Reviews (1984) and Peptides (1986). Those reviews plus Kovalzon and Strekalova (2006) give the clearest picture of what was claimed and what survived scrutiny.
Note that much of the immunoreactivity work uses antibodies whose specificity was established by the standards of the period. "DSIP-like immunoreactivity" is not the same claim as the presence of DSIP, and the distinction matters when reading the older tissue-distribution papers.
Evidence assessment
Evidence base: substantial in volume, concentrated in the 1980s, and explicitly unresolved. No specific receptor has been identified, and the reported activities have not converged on a mechanism. Treat older immunoreactivity findings with care.
